Prochlorperazine

As a commonly used antipsychotic and antiemetic drug in clinical practice, prochlorperazine is a piperazine phenothiazine derivative that exerts multiple pharmacological effects by antagonizing central dopamine D2 receptors, 5-hydroxytryptamine receptors and M1 cholinergic receptors. Its clinical application scenarios cover two types of core demands: on the one hand, it is used to control psychiatric symptoms such as excitement, agitation, hallucinations and delusions caused by acute and chronic schizophrenia and mania; on the other hand, it can effectively relieve nausea and vomiting caused by chemotherapy, radiotherapy, surgery and vestibular dysfunction. The applicable population includes adult patients with mental illnesses, people receiving tumor treatment and perioperative patients. Low-dose preparations can also be used for symptomatic treatment of severe vomiting in children under the guidance of doctors.

Publicly available data show that the global market size of antiemetic API has exceeded 2.8 billion US dollars in 2023. Among them, phenothiazine antiemetics occupy nearly 17% of the market share of antiemetic drugs in primary medical institutions due to their cost-effectiveness advantage. The original developer of prochlorperazine is GlaxoSmithKline, with the original brand name Compazine. The core compound patent expired globally in 1970. The currently marketed dosage forms of the original drug include tablets, injections and suppositories, with common specifications of 5mg tablets and 10mg/2mL injections. The relevant original dosage forms have been included in the FDA Reference Listed Drug Catalog, but have not yet been included in China's *Catalog of Reference Preparations for Chemical Drugs*.
