Eletriptan

Migraine is a common clinical primary headache type. Symptoms such as throbbing pain, nausea and photophobia accompanying the attack seriously affect patients' daily work and life, and there is an urgent clinical need for acute treatment regimens with both rapid onset of effect and good tolerability. Eletriptan belongs to the second-generation triptan anti-migraine drugs. It highly selectively activates the 5-hydroxytryptamine 1B/1D receptors in intracranial blood vessels and the trigeminal nervous system, inhibits excessive vasodilation and reduces the release of neurogenic inflammatory mediators, so as to rapidly relieve headache and accompanying symptoms. It is suitable for the acute treatment of migraine with or without aura in adults. Compared with the first-generation triptans, it has higher bioavailability and lower incidence of central adverse reactions.

The global market size of migraine medications maintains steady growth, and eletriptan accounts for approximately 12% of the triptan market share by virtue of its clinical advantages. In 2023, the global market size of eletriptan was approximately USD 210 million. With the increase in the awareness rate of migraine among patients and the rising demand for treatment, the compound annual growth rate is expected to remain at 3.2% by 2028. At present, the original research product still dominates the market. A number of generic drug enterprises in the European Union and India have been approved for marketing, while no generic drug has been approved in China yet, leaving a market gap with high development value.

The original research enterprise of eletriptan is Pfizer, with the trade name Relpax. Its core compound patent in the United States expired in 2017, and the European patent expired in 2019. The main dosage form approved for the original research product is tablet, with two specifications of 20mg and 40mg. It has been included in the FDA Reference Listed Drug Catalog and the Chinese Listed Medicines Catalog. Up to now, there is no eletriptan preparation approved for marketing in China, and there is no domestic or imported API registered in status A on the API registration platform, and relevant R&D pipelines are still in the progress stage.
