Desipramine
In the clinical treatment system for depression, tricyclic antidepressants are one of the most widely used categories in early application. Desipramine, as the active metabolite of desmethylimipramine, belongs to selective norepinephrine reuptake inhibitors. Its mechanism of action is to block the reuptake of norepinephrine by the presynaptic membrane and increase the concentration of norepinephrine in the synaptic cleft, thereby improving the core symptoms of depression. Compared with traditional tricyclic drugs, desipramine has a weaker inhibitory effect on 5-hydroxytryptamine reuptake, and has milder anticholinergic and sedation-related adverse reactions. It is mainly used clinically to treat various types of depression, especially endogenous depression and menopausal depression. Meanwhile, it can also assist in alleviating neuropathic pain and related symptoms of attention deficit hyperactivity disorder (ADHD), and the applicable population covers adults and adolescent patients who meet the indication assessment.
At present, the global desipramine market is generally in a mature and stable stage. Affected by the launch of new antidepressants, the market size has maintained in the range of 120 million to 150 million US dollars in recent years, with a compound annual growth rate of about 0.8%. The competitive landscape is dominated by generic drugs, with the original research product accounting for less than 10% of the market. North America and Europe are the main consumer markets, together accounting for more than 75% of global sales. The overall scale of the domestic market is still small at present, and desipramine has not been included in the national centralized procurement catalogue. Clinical application is still dominated by imported preparations, and there are few enterprises deploying generic drugs, leaving a certain market substitution space.
The original research enterprise of desipramine is Mallinckrodt Pharmaceuticals, and the original brand name is Norpramin. The core compound patent has expired globally in 1999. The main dosage form of the original marketed product is tablet, with specifications including 10mg, 25mg, 50mg, 75mg, 100mg and 150mg, which has been included in the FDA Reference Listed Drug Catalogue. At present, no desipramine preparation has been approved for marketing in China. There is no publicly available valid A-state registration number in the API registration database of the Center for Drug Evaluation (CDE) of the National Medical Products Administration, and relevant development is still in the preclinical or declaration stage. (Data as of October 2024, please refer to the official CDE website for the latest information)
In response to the needs of desipramine drug development and quality research, CATO can provide a full set of impurity reference standards for this API. Most products are in stock, and orders for in-stock products placed before 16:00 will be shipped on the same day. All products meet the compliance requirements of multiple regulations such as Chinese Pharmacopoeia and FDA, and can fully support pharmaceutical enterprises in impurity research, quality standard establishment and declaration-related work.
